Yes, if the negative charge (i.e the lone pair) is taking part in complete conjugation, the 2 electrons will be counted in 4n+2 pi electrons. Also you have to make sure they are inside the ring.
Conditions for aromaticity are-
  1. It should be a cyclic compound
  2. It should have complete conjugation
  3. it should have 4n+2 pi electrons
