Membranes and their constituent proteins are assembled in the endoplasmic reticulum. This organelle contains enzymes involved in lipid syntheses and as lipids are manufactured in the ER, they are inserted into organelle’s own membrane. This happens in part because lipids are too hydrophobic to dissolve into cytoplasm.
Similarly, trans-membrane proteins have enough hydrophobic surfaces so that they are also inserted into ER membrane while they are still being synthesized. Here, future membrane proteins make their way to ER membrane with help of a signal sequence in newly translated protein. Signal sequence stops translation and directs ribosomes – which are carrying unfinished proteins – to dock with ER proteins before finishing their work. Translation then recommences after signal sequence docks with ER and this takes places within ER membrane. Thus, by the time protein achieves its final form, it is already inserted into a membrane.
Proteins which will be secreted by a cell are also directed to ER during translation, where they end up in the lumen, the internal cavity where they are packaged for vesicular release from the cell. Hormones insulin and erythropoietin (EPO) are examples of vesicular proteins.
