Anode become negative due to oxidation and cathode become positive due to reduction
Eg   at anode  Zn   ---------> zn+2  + 2e (oxidation)
Here zn has tendency to go to solution from zn rod as zn ion and electron remains on zn rod so zn rod become negative.
At cathod cu +2   +2e ---------> cu
Here cu ions come from solution to cu rod and neutralise to getting 2 electron from cu rod, so it become positive.

This condition is achieved after inserting u tube....I'm just asking you before that what will be the anode and cathode
