  • Wavelength of Light: Shorter wavelengths provide better resolution. For example, blue light has a shorter wavelength than red light, leading to improved detail.
  • Numerical Aperture (NA): This is a measure of the lens's ability to gather light and resolve fine specimen detail. A higher NA results in better resolving power.
  • Medium Between Lens and Specimen: The refractive index of the medium (like air or oil) can enhance resolution. Immersion oils are often used to increase NA.

Expression of Resolving Power

The resolving power (R) can be mathematically expressed using the formula:

R = λ / (2 * NA)

Here, λ represents the wavelength of light used, and NA is the numerical aperture of the lens. This equation highlights how both the wavelength and the lens's capability work together to determine the microscope's resolving power.
