Procedure:
Prepare the Leaf:

Place the leaf in a cup or a beaker.
Boil the Leaf:
Pour boiling water over the leaf to soften it and to stop any enzymatic activity that could interfere with the test.
Allow the leaf to remain in the boiling water for about 2-3 minutes.
Remove the Leaf:
After boiling, carefully remove the leaf from the water using forceps or tweezers and place it into a dish of ethanol.
Leave the leaf in the ethanol for a few minutes. This step helps to remove the chlorophyll from the leaf.
Prepare Iodine Solution:
While the leaf is soaking in ethanol, prepare the iodine solution if you haven't already. Dilute the iodine solution with water if it's too concentrated.
Stain the Leaf:
After the leaf has been in ethanol for a few minutes, remove it from the ethanol and rinse it with water.
Now, place the leaf on a white plate or a Petri dish.
Using a dropper or a pipette, carefully apply a few drops of iodine solution onto the leaf. Make sure to cover the entire surface of the leaf with the solution.
Observe:
Wait for a few minutes and observe the leaf.
If starch is present in the leaf, it will turn blue-black in color where the iodine solution has been applied. This color change indicates the presence of starch.
Interpretation:
Blue-black coloration indicates the presence of starch.
If there's no color change, or the leaf remains brownish, it means there's no starch present.

Make sure to handle the iodine solution carefully, as it can stain skin and clothes.
Take precautions while using boiling water and ethanol to avoid accidents.
