Chance of eating pizza!\u00c3\u201a\u00c2\u00a0 No chance; they are going to leave.<\/p>\n<p style=\"text-align: justify;\">I believe you are one of them. Did you ever think; pizza that you are eating; how its base is prepared?<\/p>\n<p style=\"text-align: justify;\">Well, certain microorganisms are used for making such products. We had been using these techniques of consuming microorganisms for our benefits from the age-old time. \u00c3\u201a\u00c2\u00a0We will better call it as <b>old biotechnology.\u00c3\u201a\u00c2\u00a0 <\/b>\u00c3\u201a\u00c2\u00a0<b>Old Biotechnology<\/b> includes the techniques which are based upon the intrinsic capacity of microorganisms which help in obtaining certain product. Manipulation is done at cellular\/genetic level. It may involve the use of algae, fungi, bacteria, yeast, cells of animals &amp; plants etc.<\/p>\n<p style=\"text-align: justify;\">These microorganisms were more exploited for more production which spearheaded towards the concept of <b>Modern Biotechnology<\/b>.<\/p>\n<p style=\"text-align: justify;\"><b>Biotechnology<\/b> in India is on a sturdy growth. In cultivating global profile of any country, indeed biotechnology plays an immense role. \u00c3\u201a\u00c2\u00a0One won\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u201e\u00a2t deny that biotechnology is the driving cause for several industries like chemical, drug, medicine and food. India stakes <b>2%<\/b> of the world\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u201e\u00a2s biotech industry. There are around <b>800 biotech companies<\/b> in India which make it an emerging bio service provider.<\/p>\n<p style=\"text-align: justify;\">We are already aware of the thing, that India is second most populated country of globe. To overcome the food consumption and demand of mankind, one has to step forward towards the technology.<\/p>\n<figure id=\"attachment_8844\" aria-describedby=\"caption-attachment-8844\" style=\"width: 346px\" class=\"wp-caption aligncenter\"><a href=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/2.jpg\"><img decoding=\"async\" class=\"size-full wp-image-8844\" src=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/2.jpg\" alt=\"\" width=\"346\" height=\"346\" \/><\/a><figcaption id=\"caption-attachment-8844\" class=\"wp-caption-text\">Fig B: Alexander Fleming<\/figcaption><\/figure>\n<p style=\"text-align: justify;\">In 1928<b>, Alexander Fleming (Fig B) <\/b>did a significant discovery of Penicillin.\u00c3\u201a\u00c2\u00a0Later it was found to be having a figure of antibiotic properties. <b><br \/>\n<\/b><\/p>\n<p style=\"text-align: justify;\">Production of <b>recombinant insulin (1975)<\/b> was a revolutionary phase in biotechnology. \u00c3\u201a\u00c2\u00a0Let\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u201e\u00a2s move the cursor for getting more information about Insulin.<\/p>\n<p style=\"text-align: justify;\">Insulin is a protein hormone which is produced in Pancreas &amp; it consists of two peptide chains viz. A &amp; B chain (Fig C). It helps in <b>decreasing <\/b>the<b> blood<\/b> <b>sugar level<\/b> (Normal Blood Sugar \u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u20ac\u0153 around 100 mg\/dL).<\/p>\n<figure id=\"attachment_8845\" aria-describedby=\"caption-attachment-8845\" style=\"width: 689px\" class=\"wp-caption aligncenter\"><a href=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/3.png\"><img decoding=\"async\" class=\" wp-image-8845\" src=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/3.png\" alt=\"3\" width=\"689\" height=\"441\" \/><\/a><figcaption id=\"caption-attachment-8845\" class=\"wp-caption-text\">Structure of insulin<\/figcaption><\/figure>\n<p style=\"text-align: justify;\">Initially, <b>bovine insulin<\/b> (1922) used to be given to patients by injection. \u00c3\u201a\u00c2\u00a0An impure form of insulin was injected to <b>Leonard Thompson<\/b>. A callus was formed at the injection site (left buttock). It was not found to be suitable then. The purification work was then continued. \u00c3\u201a\u00c2\u00a0Though chromatographic purification techniques (1974), highly purified animal insulin was produced. Fully synthetic insulin was produced in 1975 in laboratory of <b>Ciba-Geigy in Base. <\/b>Its clinical trial was also successful on patients. This significant invention of <b>recombinant DNA technology <\/b>helped in treating Diabetic patients.<\/p>\n<p style=\"text-align: justify;\"><b>Recombinant DNA technology! What is that?<\/b><\/p>\n<p style=\"text-align: justify;\">It includes a series of procedures in which two DNA segments are recombined. This recombined DNA replicates in a cell using host machinery. Simply saying, it integrates itself into that cell.<\/p>\n<p style=\"text-align: justify;\">Different tools and techniques are used in <b>recombinant DNA technology.<\/b> \u00c3\u201a\u00c2\u00a0Let\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u201e\u00a2s take a look to the <b>tools <\/b>of this technology first.<\/p>\n<p style=\"text-align: justify;\"><b>1.\u00c3\u201a\u00c2\u00a0\u00c3\u201a\u00c2\u00a0\u00c3\u201a\u00c2\u00a0\u00c3\u201a\u00c2\u00a0\u00c3\u201a\u00c2\u00a0\u00c3\u201a\u00c2\u00a0 <\/b><b>Enzymes<\/b><\/p>\n<ul style=\"text-align: justify;\">\n<li><b>Cleaving enzyme<\/b> \u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u20ac\u0153<b>Restriction endonuclease<\/b> (Molecular scissors) (Fig D) enzyme cleave DNA duplex at specific points. That localized specific points are known as <b>recognition sit<\/b>e or <b>restriction site. <\/b>These enzymes cleave in such a manner that single stranded free ends project from each fragment of DNA duplex.\u00c3\u201a\u00c2\u00a0 These single stranded free ends are called \u00c3\u00a2\u00e2\u201a\u00ac\u00cb\u0153sticky ends\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u201e\u00a2 because they can join related complimentary ends of DNA fragment from some other source.<\/li>\n<\/ul>\n<figure id=\"attachment_8846\" aria-describedby=\"caption-attachment-8846\" style=\"width: 361px\" class=\"wp-caption aligncenter\"><a href=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/4.gif\"><img loading=\"lazy\" decoding=\"async\" class=\"size-full wp-image-8846\" src=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/4.gif\" alt=\"\" width=\"361\" height=\"217\" \/><\/a><figcaption id=\"caption-attachment-8846\" class=\"wp-caption-text\">Fig D : Restriction Enzyme (EcoRI)<\/figcaption><\/figure>\n<ul style=\"text-align: justify;\">\n<li><b>Joining enzyme<\/b> (Molecular Glues) Fig E\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u20ac\u0153 These enzyme help in joining the DNA fragment. For example DNA ligase from Escherichia coli is used to join DNA fragments. Cloning Vector\u00c3\u201a\u00c2\u00a0<\/strong><\/p>\n<p style=\"text-align: justify;\">It is the vehicle or carrier DNA which is used for transferring a DNA fragment into host cell. \u00c3\u201a\u00c2\u00a0Some of the cloning vectors are:<\/p>\n<ul style=\"text-align: justify;\">\n<li><b>Plasmids <\/b>\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u20ac\u0153 Plasmids are extra chromosomal genetic material. They are basically loops of DNA. Plasmids were discovered by <b>William Hays &amp; Joshua Lederberg<\/b> (1952) in bacterial cell. A bacteria cell may hold one or many copies of plasmids (Fig E).<\/li>\n<\/ul>\n<p style=\"text-align: justify;\">There are many varieties of plasmids which differ from one another in size &amp; in composition of genes. They vary in size from 2 Kilo base pairs to 400 Kilo base pairs.<\/p>\n<p style=\"text-align: justify;\">You know the major component of plasmid comprise of an <b>origin of replication<\/b> which act as a start signal for self-replication. The important components of a plasmid include (Fig F) antibiotic resistant region, specific restriction site &amp; origin of replication.<\/p>\n<p style=\"text-align: justify;\">Because of antibiotic resistant region, plasmids are not killed by antibiotic. They are able to survive and grow in presence of antibiotic. E.g. <b>pBR322<\/b>, which is a cloning vector obtained from <b><i>E.Coli<\/i><\/b> bacteria. Discovery of bacteriophage was done by two scientists Frederick W. Twort\u00c3\u201a\u00c2\u00a0 (Britain, 1915) &amp; Felix d Herelle (France, 1917). Bacteriophages are contributing as a significant part of Modern Biotechnology.<\/li>\n<\/ul>\n<figure id=\"attachment_8851\" aria-describedby=\"caption-attachment-8851\" style=\"width: 360px\" class=\"wp-caption aligncenter\"><a href=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/9.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\" wp-image-8851\" src=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/9.jpg\" alt=\"9\" width=\"360\" height=\"346\" \/><\/a><figcaption id=\"caption-attachment-8851\" class=\"wp-caption-text\">Fig G: Structure of bacteriophage<\/figcaption><\/figure>\n<ul style=\"text-align: justify;\">\n<li><b>Cosmids <\/b>\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u20ac\u0153 They are the hybrid plasmids. They are effectively used for transferring the DNA to host.<\/li>\n<li><b>Phagemids<\/b> \u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u20ac\u0153 it consists of origin of replication from filamentous phage \u00c3\u00a2\u00e2\u201a\u00ac\u00cb\u0153f1\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u201e\u00a2. It is combined with M13 phage.<\/li>\n<li>Other than this <b>BACs <\/b>(Bacterial artificial chromosomes) &amp; <b>YACs<\/b> (Yeast artificial chromosomes) are also used as vectors.<\/li>\n<\/ul>\n<p style=\"text-align: justify;\"><b>1.\u00c3\u201a\u00c2\u00a0<\/b><b>Passenger DNA<\/b><\/p>\n<p style=\"text-align: justify;\">This DNA is to be cleaved and to be cloned. It is <b>the foreign DNA<\/b>. It can either be synthetic DNA, random DNA or copied DNA.<\/p>\n<p style=\"text-align: justify;\"><b>2.\u00c3\u201a\u00c2\u00a0\u00c3\u201a\u00c2\u00a0<\/b><b>Host cell<\/b><\/p>\n<p style=\"text-align: justify;\">It is the cell in which recombinant molecule (Passenger DNA + vector) is going to reproduce. It should be competent enough to take up the new DNA. We can also increase its competency. How? I<\/strong><b>solation of genetic material<br \/>\n<\/b>For starting with the DNA work, first thing we have to do is isolation of DNA from cell. How we are going to do that?<\/p>\n<p style=\"text-align: justify;\">DNA is present inside the nucleus and for separating it we need to break cell membrane of cell and then for nucleus. This is known as <b>cell lysis<\/b>. Cell lysis is done by a number of methods like enzymatic reaction, detergent, blending etc.<\/p>\n<p style=\"text-align: justify;\"><b>2.\u00c3\u201a\u00c2\u00a0<\/b><b>Cutting of DNA of specific location<\/b><\/p>\n<p style=\"text-align: justify;\">Gene of interest is isolated from the \u00c3\u00a2\u00e2\u201a\u00ac\u00cb\u0153donor DNA\u00c3\u00a2\u00e2\u201a\u00ac\u00e2\u201e\u00a2. Amplification of Gene of interest using PCR:<\/b><\/p>\n<figure id=\"attachment_8854\" aria-describedby=\"caption-attachment-8854\" style=\"width: 717px\" class=\"wp-caption aligncenter\"><a href=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/141.png\"><img loading=\"lazy\" decoding=\"async\" class=\" wp-image-8854\" src=\"https:\/\/www.askiitians.com\/blog\/wp-content\/uploads\/2016\/03\/141-1024x534.png\" alt=\"14\" width=\"717\" height=\"374\" \/><\/a><figcaption id=\"caption-attachment-8854\" class=\"wp-caption-text\">Fig J: Polymerase chain reaction<\/figcaption><\/figure>\n<p style=\"text-align: justify;\">DNA sample obtained after cutting with restriction enzyme is amplified using polymerase chain reaction (Fig J). Picture shows formation of multiple copies of DNA. It is done in three steps. Initially DNA strands are separated by increasing the temperature (<b>denaturation<\/b>). Primers; which are an oligonucleotide strand are added &amp; they bind to its complementary sequence on DNA.\u00c3\u201a\u00c2\u00a0 Temperature is also lowered then (<b>annealing<\/b>). DNA polymerase enzyme adds nucleotides to the primers &amp; extends the DNA sequence (<b>elongation<\/b>).<\/p>\n<p style=\"text-align: justify;\"><strong>4<\/strong><b>.<\/b><b>Insertion of recombinant DNA into host<\/b><\/p>\n<p style=\"text-align: justify;\">Foreign DNA is introduced into host cell. It is known as<b> transformation. \u00c3\u201a\u00c2\u00a0<\/b>Transformation itself is done by a number of techniques. It will take two more pages to write that story. Obtaining foreign Gene product<br \/>\n<\/b>Why we are doing so much of stuff. Obviously! For obtaining the desired protein. All favorable conditions have to be provided to host for better expression of recombinant DNA.
