Protein is an amino acid sequence, but an amino acid sequence is called protein only when it is responsible for any character. Not all amino acid sequence is responsible for characteristics.
Even though the possibilities are millions but only few could be functional.

The number of distinct proteins containing n amino acids is 20^n. 
Hence total nos. of proteins are 20^100.
